14:26-27 What is the significance of this passage?

Here we see the return of Paul and Barnabas to Antioch in Syria at the completion of their first apostolic mission journey, which began in Ac 13:1 (CP Ac 13:1-4).

This apostolic mission journey, taking approximately three years to complete, took Paul and Barnabas first to the island of Cyprus, then across to Asia Minor where they founded churches at Antioch in Pisidia, and Iconium, Lystra and Derbe in Galatia (CP 13:14-16, 43-48; 14:1-7, 19-23).

It was at Lystra that a young man named Timothy became one of Paul's converts. Paul became Timothy's mentor, teaching and encouraging him in many areas of ministry until God called him to be an apostle also (CP 16:1-5; 1Ti 1:1-2, 18 with 1Th 1:1, 2:1-6).
