Perfected in this context means to complete, finish a work or duty, bring to an end by reaching the intended goal.
The idea was that Jesus was going to work that day and the next, and then bring His earthly work to an end on the third day when He would die. Jesus said this on the way to Jerusalem (cp V22).
Jesus was not concerned that Herod would kill Him on the way because He was appointed to die in Jerusalem (cp Mk 10:33-34). See also comments on Lk 9:51.
